Cells in our bodies are constantly subjected to mechanical forces, which impact cellular decision-making (differential gene expression) in development and disease. The Alushin lab is investigating how forces modulate the conformation of macromolecular assemblies, in particular cytoskeletal filaments, to regulate binding interactions which transduce mechanical cues into chemical signaling pathways. We are developing technology for elucidating the structures of macromolecules under load with cryo-electron microscopy, as well as employing fluorescence-based methods to visualize the dynamics of these processes in vitro and in cells.
